Description
The principle of BCA protein quantification method is that under
alkaline conditions, the peptide bond in the protein can reduce Cu2+
to Cu+ (biuret reaction), and the reduction amount of Cu2+ is
proportional to the total protein amount, and the generated Cu+ can
form with BCA. Stable purple compound. Compared with the Bradford
method, the BCAprotein quantification method is more objective. The
common peptide bond can produce color reaction, and it is also
compatible with high concentration SDS (5%), Triton X-100 (5%), NP 40
(5%) , Tween 20, 60, 80 (5%), its disadvantage is that it is susceptible to
certain reducing agents (such as dithiothreitol and β-thioethanol,
etc.), chelating agents (EDTA, EGTA) and high concentration buffers ,
but these interferences can be eliminated by diluting the protein
samples.

Interfering Substances

those substances with reducing potential, chelating agents, and strong
acids/bases. The following table shows the highest concentration of
these substances in the protein sample buffer without interfering with
the BCAassay.
